Na vroege onderzoeksposities in het St. Luke's Hospital in Detroit en New York, vond Kendall zijn plaats bij de Mayo Foundation (een deel van de Universiteit van Minnesota) in Rochester, Minnesota, waar hij zijn onderzoek eerst op de schildklier concentreerde. In december 1914 bereikte hij een baanbrekende ontdekking toen hij het schildklierhormoon isoleerde, later thyroxine genoemd. Toen thyroxine met succes werd gesynthetiseerd, meer dan een decennium later, werd het een effectieve behandeling voor schildklierdeficiënties.

Kendall ging vervolgens over tot het bestuderen van de hormonen van de bijnierschors. Hoewel men geloofde dat de bijnieren slechts één hormoon afscheiden, bewees Kendall in 1939 dat er in werkelijkheid meerdere waren. Hij bracht het volgende decennium door met het onderzoeken van deze hormonen, in het bijzonder een die hij in eerste instantie verbinding E noemde. Tegen 1949 had Kendall het hormoon bewezen dat effectief was bij het verlichten van reumatoïde artritis. Het jaar daarop ontving hij met zijn onderzoekscollega Philip S.
